چکیده

Recent work has identified ethanol toxicity as a major factor pre venting continuous production of ethanol at the concentrations ob tainable in batch culture. In this paper we investigate the use of a con tinuous two-stage bioreactor design to circumvent toxic effects of ethanol. Biomass is produced via continuous culture in the first stage reactor in which ethanol concentrations are either absent or main tained at low levels. The freshly grown cells are fed into the second bioreactor in which high ethanol concentrations are produced. The steady influx of fresh cells and continuous removal of spent cells helps minimize the loss of fermentative activity that results from anaerobiosis and exposure to high ethanol concentrations. A final ethanol concentration of 37 g L-1 and overall yield of .32 g g-1 were obtained with the two-stage reactor as compared to corresponding values of 38 g L-1 and .32 g g-1 obtained in batch. The volumetric rate in the two-state process was .96 g L-1 as compared to .46 g L-1h-1 in batch. *Author to whom all correspondence and reprint requests should be addressed. †Maintained in cooperation with the University of Wisconsin-Madison. Continuous ethanol production from D-xylose by Candida shehatae.

Much progress has been made since the discovery that some yeasts can ferment D-xylose to ethanol. 1 Initially, only a few yeasts were found that could produce ethanol from D-xylose. Today, at least eight species of yeasts are known to produce significant amounts of ethanol from D-xylose. 2,4-6 Three of these species have been studied extensively: Pachysolen tannophilus, Candida shehatae, and Pi...

Effects of Culture Conditions on the Fermentation of Xylose to Ethanol by Candida shehatae

This research examined four factors on the fermentation of xylose by Candida shehatae, and the following conclusions were reached: (1) A minimal medium is effective for producing ethanol. (2) Peptone and casamino acids stimulate ethanol production. (3) Aeration is important inobtaining good ethanol production rates and yields. (4) The maximal rate ofethanol production is attained around pH 3.2-...

Purification and Properties of Xylitol Dehydrogenase from the Xylose-fermenting Yeast Candida shehatae

Xylitol dehydrogenase (EC1.1.1.9) from xylose-grown cells of Candida shehatae was purified 215-fold by sequential chromatography on NADC8 affinity, Superose-12, and Cibacron blue columns, and a single band was observed by SDS gel electrophoresis. The purified enzyme had a native molecular weight of 82 kDa and a denatured molecular weight of 40 kDa following SDS gel electrophoresis, indicating t...

The influence of pH and aeration rate on the fermentation of D-xylose by Candida shehatae.

The effects of the initial pH and air supply on the production of ethanol from D-xylose using the yeast Candida shehatae in a batch reactor were investigated. The initial pH was altered within the range of 2.5-6.5 and the specific aeration rate from 0.0-0.3 vv-1 min-1. The results showed that the most favorable initial pH for ethanol production was 4.5 and aeration via the stirring vortex of th...

Evaluation of novel xylose-fermenting yeast strains from Brazilian forests for hemicellulosic ethanol production from sugarcane bagasse

Bioconversion of hemicellulosic hydrolysates into ethanol with the desired yields plays a pivotal role for the overall success of biorefineries. This paper aims to evaluate the ethanol production potential of four native strains of Scheffersomyces shehatae (syn. Candida shehatae) viz. S. shehatae BR6-2AI, CG8-8BY, PT1-1BASP and BR6-2AY, isolated from Brazilian forests. These strains were grown ...
